ABSTRACT
A macromolecular crystallography beamline BL41XU allows for diffraction experiment by using the photon flux of the 2.2 × 1012 〜 1.1 × 1013 (photons/s@1 Å) which is the highest level in the world. The beamline is mainly used for the structural determination of challenging targets such as membrane proteins and macromolecular complexes which are usually difficult to make well diffracting crystals. Since the beam size can be changed from 4 × 5 to 22(H) × 45(V) µm2, it can cope with various samples whose size is from a few micrometer to several hundreds micrometer. Another characteristic feature of BL41XU is that it allows for data collection using high energy X-ray from 19 keV to 35 keV.

SOURCE AND OPTICS
The light source of BL41XU is the SPring-8 standard in-vacuum undulator. A liquid –nitrogen-cooled Si double crystal monochromator is used to monochromatize the beam. For the normal mode that covers from 6.5 to17.7 keV the horizontal two step focusing optics consisting of the first horizontal mirror and KB mirror is installed. For the high energy mode that covers from 19 to 35 keV compound refractive lens is used for beam focus.
Energy (wavelength) | Normal mode: 6.5 - 17.7 keV (1.9 - 0.7 Å) High energy mode†: 19 - 35 keV (0.65 - 0.35 Å) |
Energy resolution | ΔE/E < 2 × 10-4 |
Beam divergence | < 1 mrad |
Beam size | Normal mode: 2 × 2 µm2 - 35 (H) µm × 50 (V) µm2 (routine use‡: 4(H) × 5(V) µm2 - 22(H) × 45(V) µm2) High energy mode: 30 × 30 µm2 - 300 × 300 µm2 |
Photon flux | Normal mode: 1.7 × 1012 – 4.3 × 1013 (phs/sec@12.4 keV) (routine use‡: 2.2 × 1012 ~ 1.1 × 1013 (phs/sec) ) High energy mode: 1.9 × 1010 〜 2.6 × 1012 (phs/sec@30 keV) |
†Users who wish to use the high-energy mode should contact the beamline scientist before applying for beamtime.
‡Users who wish to use the beam size and photon flux other than those should contact the beamline scientist before applying for beamtime.
